Brief Introduction:

 

Doxycycline hyclate is a semi-synthetic tetracycline antibiotic that can penetrate cell membranes and bind to 30S ribosome subunits, interfering with tRNA/mRNA interactions and ultimately inhibiting protein synthesis. As a broad-spectrum antibiotic and bacteriostasis agent, this antibiotic has very strong resistance mainly to the bacteria that cause respiratory infections including Mycoplasma pneumoniae and Haemophilus influenzae. In addition, it can also treat chlamydia, rickettsii and some spirochete caused by infection and has antigenic and repellant function.

 

Doxycycline hydrochloride is also an inhibitor of several matrix metalloproteinases (MMPs), including MMP-8 (neutrophil Collagenase; IC50 =30 μM) and MMP-1 (Interstitial Collagenase; IC50 = 300 µM). It is reported that it can significantly reduce the expression and activity of MMP-9 in recurrent corneal erosion. In smooth muscle studies, it can enhance and promote smooth muscle cell adhesion, change fibrous collagen matrix reorganization and cell migration. It also inhibits inflammation caused by Lyme Disease and interferes with the plasminogen system to protect microvessels.

 

Doxycycline hydrochloride is also commonly used in teT-ON (RTTA-dependent) and TET-OFF (TTA-dependent) systems to regulate gene expression, both in vivo and in vitro cell cultures. This product is a USP grade reagent with a solubility of approximately 50 mg/mL in water and heated moderately to promote complete dissolution.According to the literature, it can also dissolve in DMSO (~1 mg/mL) and methanol, insoluble in chloroform and ether.
